The other name of the Church, given after its high altar, is the Transfiguration Church in Bolshaya Ordinka (or "in Ordintsakh"). It was first mentioned in the 1571 chronicle, when the Church was dedicated to St. Barlaam of Khutyn. The Church was founded in the time of Metropolitan Barlaam, in 1523. The altar of the Transfiguration has been known since 1625.
The stone church was built from 1683 to 1685.
After the Church's Joy of All Who Sorrow Icon of Our Lady was glorified in 1688, the Church got the name of this wonder-working icon.
In the late 18th century, the western side of the Church with a three-tier bell tower was rearranged by architect Vasili Bazhenov.
The Church was damaged during the 1812 fire. From 1831 to 1836, it was reconstructed by architect Joseph Bove with support from the brothers Kumanin and A. Dolgov.
The Church was shut down in 1930. The bells were taken off and destroyed. During World War II, there was a depositary of the State Tretyakov Gallery in the Church. As a result, the interior furnishings remained safe and intact. The Church was consecrated again in 1948.
